Probiotics are living inputs: every extra minute of heat, shear, moisture or oxygen exposure in a larger vessel removes viable cells before the finished unit is even packed. When that loss is not modelled at pilot stage, the shortfall shows up at release testing, and it is paid for through heavier overage, re-quoted pricing, postponed launch dates or a quietly revised label claim. Assessing scale-up capability before you approve a formula is far cheaper than renegotiating after a batch fails.

Small-batch samples and commercial batches differ in three variables that matter most to viability: mixing, temperature control and exposure time. A bench blender reaches uniformity in a minute or two; a production ribbon or ploughshare mixer may need considerably longer to distribute a low-dose culture, and every extra minute adds shear and frictional heat. Small masses shed heat quickly, while large masses hold it, so jacket temperature setpoints that worked in a pilot vessel no longer describe what the powder actually experiences. Larger batches also stand longer, in hoppers, in open transfers and in drums waiting for the next step. Surface-to-volume ratio falls as volume grows, so cooling and drying slow down and mixing dead spots become more likely.

Viability damage accumulates rather than resets. A culture carries the thermal history of fermentation, concentration and either freeze-drying or spray drying before it ever reaches the blending room; the factory then adds blending time, compression or cooking heat, and filling-room exposure on top. Spray drying exposes cells to high inlet temperatures for a short residence time, while freeze-drying is gentler but leaves residual moisture and matrix formulation to determine what happens next. If a manufacturer buys concentrated cultures from a third party, it inherits that history and adds its own. The relevant question is not the incoming CFU per gram on the certificate, but the cumulative exposure from that certificate to the sealed unit.

A sample proves a factory can make the product once. A validated scale-up record proves it can make it every time, at the volume you are actually buying.

Tableting is usually the harshest dosage step for live cultures. Compression generates local heat and pressure at the punch face, and both rise with force, with deep concave tooling, with low-porosity formulations and with faster turret speeds that shorten dwell time. Chewable formats often need gentler compression, which shifts the burden onto binder and filler selection. Gummies present a different problem: the culture has to enter a hot, high-moisture matrix, and the cooling window decides how long it stays there. In pilot moulds, gummies set quickly; on a wide production line with deep trays, the centre of the piece may stay warm far longer, and that extended window is where post-addition losses accumulate.

Stick sachets and powders fail in a quieter way. A probiotic is usually a very small fraction of the formula, so blend uniformity depends on geometric dilution, carrier particle size matching and a mixer that does not segregate the batch when it is discharged into a hopper. Filling accuracy matters just as much: at low fill weights, small absolute deviations translate into wide relative variance between sachets, so the batch mean has to be pushed higher to keep the low end of the distribution above label claim, and that overage is a direct cost. Capsule filling brings humidity into play. Hygroscopic powder picks up moisture in the hopper, sticks, and carries free water into the capsule, where it shortens survival during storage. Room dew point, hopper residence time and whether capsules are banded or sealed belong in the technical agreement.

Setting the CFU target is where scale-up data becomes commercial leverage. The label claim should be built from measured step yields rather than from a habitual overage: incoming blend potency, loss across blending, loss across compression, cooking or filling, loss to release, and loss across shelf life. Once each step has a number, the overage is defensible and quotes become comparable between suppliers. Water activity and product temperature are the two process parameters most worth controlling explicitly, with limits set from trials rather than copied from a template, because both drive the rate at which remaining viable cells die. In-process sampling should cover the blended powder, the middle and end of the run, and the finished unit, and three consecutive validation batches at commercial scale are what turn those samples into a batch-to-batch consistency claim that stability testing can then confirm.

Before you approve a sample, ask the factory a short set of questions and expect documents, not assurances. Has this exact dosage form been run commercially in the last year or two, and can they share step yields from those batches? Were the validation batches made at commercial scale and speed, or on a pilot line that only resembles production? If release CFU falls below specification, who carries the loss, what counts as a rework versus a reject, and what are the release criteria beyond CFU, including water activity and content uniformity? Finally, how is the CFU guarantee written into the quote and the lead time, since a higher overage needs more concentrate and longer culture procurement, and that has to be visible before you commit to a launch date.

The practical rule is to treat the pilot as a scale-up exercise rather than a beauty sample. Ask for production-intent parameters, request the loss data behind the overage, and choose a partner whose commercial record covers the format you are buying. A factory that can explain where CFU disappears in its own process is usually also the one that can hold the claim on your label.
","CFU loss, batch drift and dose unevenness between
